Catalytic Triad His57 Asp102 Ser195 H H O N C [HOOC] H O [NH2] Chymotrypsin 的作用機制分成兩個階段,每個階段各有數個步驟。其詳細說明請見講義或課本,同時網頁上有動畫說明,請仔細觀察琢磨。 在此可對胜肽鍵的水解做一整理,找出一些共同的機制。大致上都是胜肽鍵上的 carbonyl 碳 (C=O) 被具有強烈負電基團的 -O: (或者水分子上面的氧原子) 攻擊,產生以碳為中心的過渡狀態,然後再斷掉胜肽鍵。Chymotrypsin 的順序式水解則含有兩套如此的攻擊事件,分別由 (1) 活性 Ser 基團上的 -O: 及 (2) 水分子擔任攻擊手。 Check substrate specificity E5-18
