Pengecatan ini termasuk pengecatan positif-diferensial. Cuplikan inokulum harus diletakkan dalam gelas benda dan difiksasi dengan pemanasan. Beberapa reagen yang diperlukan adalah: Crystal Violet (Primary Stain) Iodine (Mordant) Decolorizer (ethanol) Safranin (Counterstain) Aquades (dalam botol semprot)
